PLEASE HELP How to remove a trojan virus (downloader)?

My antivirus software (Trend Micro for Internet security) is to date, but not clean, delete or quarantine the virus. Microsoft Deleter malware does not work. Windows Defender does not work on it and not let me delete file physically well. I need to know how to destroy this virus, because it keeps opening pages and slowing down my computer. Besides, I do not I have nothing screwing with my files because the year 12 of the instant messaging company yet. PLEASE HELP
